Nieuwe flowcytometers met toenemend aantal fluorescerende kanalen in de UV en Far Red en de
beschikbaarheid van overeenkomstige fluorescerende conjugaten zijn nu beschikbaar. Om bij te blijven, hebben we de Ultra Rainbow Fluorescent Particles ontwikkeld met verbeterde UV- en Far Red-fluorescentie-intensiteit. De Ultra Rainbow Fluorescent Particles bevatten een enkele piek en zijn ontworpen voor het controleren van de optische uitlijning van elke flowcytometer in alle kanalen.
Beschrijving
Dit product bevat een enkele populatie regenboogdeeltjes die zijn geverfd tot een enkele fluorescerende intensiteit. Elk regenboogdeeltje bevat een mengsel van fluoroforen die stabiel zijn ingebed in het deeltje en kunnen worden geëxciteerd bij elke golflengte van 365-650 nm, waardoor de meeste kanalen in een flowcytometer kunnen worden gekalibreerd met dezelfde set deeltjes. De Rainbow Particles hebben emissiespectra die compatibel zijn met veel gangbare fluoroforen die worden gebruikt voor immunofluorescentiekleuring door middel van flowcytometrische analyse. Rainbow Fluorescent Particles worden geleverd in een druppelflesje voor gemakkelijke dosering en opslag.
Deze Rainbow Calibration Particles zijn een mengsel van 3,0 – 3,4 m deeltjes, gekleurd tot zes verschillende fluorescerende intensiteiten. Omdat hun excitatiebereik 365 – 650 nm is, kunnen ze met de meeste lasers worden gebruikt. Ze zijn echter niet voor gebruik met de UV-laser. De emissiespectra van deze regenboogdeeltjes zijn compatibel met veel gebruikelijke fluoroforen die worden gebruikt bij immunofluorescentiekleuring met flowcytometrische analyse.
Aanbevolen testprocedures
Dit deeltjesmengsel (~10×10^6 deeltjes/ml) is nuttig voor routinematige kalibratie van flowcytometers. Resuspendeer de deeltjes vóór gebruik door ze te vortexen. Verdunning van 3-5 druppels deeltjes tot 1 ml omhulselvloeistof zal een voldoende aantal deeltjes opleveren voor flowcytometrische analyse.

- Let op: Natriumazide levert onder zure omstandigheden zeer toxisch hydrazolzuur op. Verdun azideverbindingen in stromend water voordat u ze weggooit om ophoping van mogelijk explosieve afzettingen in leidingen te voorkomen.
