Lipideperoxidatie is een bekend voorbeeld van oxidatieve schade in celmembranen, lipoproteïnen en andere lipide-bevattende structuren. Peroxidatieve modificatie van onverzadigde fosfolipiden, glycolipiden en cholesterol kan in verschillende reacties optreden. Ze kunnen worden geactiveerd door i) vrije radicalen zoals oxylradicalen, peroxylradicalen en hydroxylradicalen afgeleid van ijzer-gemedieerde reductie van waterstofperoxide of ii) niet-radicalen zoals singlet zuurstof, ozon en peroxynitriet gegenereerd door de reactie van superoxide met stikstofmonoxide.

Malondialdehyde (MDA) en 4-hydroxyalkenalen zijn belangrijke giftige bijproducten van lipideperoxidatie. De meting van de hoeveelheden van dergelijke aldehyden komt dus overeen met een index van lipideperoxidatie  in vitro  en  in vivo . 4-Hydroxynonenal (4-HNE) is een belangrijk product van de peroxidatieve afbraak van ω-6 meervoudig onverzadigde vetzuren (PUFA). Het bezit cytotoxische, hepatotoxische, mutagene en genotoxische eigenschappen. Verder werden verhoogde niveaus van HNE gevonden in plasma en verschillende organen onder omstandigheden van oxidatieve stress. In feite is MDA in veel gevallen het meest voorkomende individuele aldehyde als gevolg van lipideperoxidatie. In vitro  kan MDA eiwitten, DNA, RNA en vele andere biomoleculen veranderen.

De LPO- assaykit van Bioquochem   meet MDA- en HNE-concentraties als een index van lipideperoxidatie. Ten eerste initieert zuur-gekatalyseerde aanval op de 3-positie van de indolring de reacties tussen indolen en aldehyden (MDA en HNE). Als resultaat geeft deze reactie een diindolylalkaan (chromofoor) met een maximale absorptie in het gebied van 580-620 nm.

In onze test reageert een indol (reagens A) snel met MDA en HNE in zuur medium, wat een chromofoor (C) oplevert met een hoge molaire extinctiecoëfficiënt bij zijn maximale absorptiegolflengte van 586 nm.

 

  • Productoverzicht

    Lipideperoxidatie (MDA) Assay Kit (colorimetrisch/fluorometrisch) (ab118970) biedt een handig hulpmiddel voor gevoelige detectie van malondialdehyde (MDA).

    In het lipideperoxidatietestprotocol reageert het MDA in het monster met thiobarbituurzuur (TBA) om een ​​MDA-TBA-adduct te genereren. Het MDA-TBA-adduct kan gemakkelijk colorimetrisch (OD = 532 nm) of fluorometrisch (Ex/Em = 532/553 nm) worden gekwantificeerd. Deze test detecteert MDA-niveaus zo laag als 1 nmol/well colorimetrisch en 0,1 nmol/well fluorometrisch.

    De MDA-assay wordt ook wel een TBARS-assay genoemd.

    Samenvatting van protocol voor lipideperoxidatie-assay:
    – voeg TBA-oplossing toe aan monsters en standaarden, incubeer gedurende 60 minuten bij 95ºC, koel gedurende 10 minuten in ijsbad
    – breng over naar putjes van de microtiterplaat
    – analyseer met microtiterplaatlezer
    Voor hogere gevoeligheid, precipiteer met n-butanol, centrifugeer, droog en resuspendeer de pellet vóór analyse.

  • Opmerkingen:

    Lipideperoxidatie verwijst naar de oxidatieve afbraak van lipiden. In dit proces nemen vrije radicalen elektronen uit de lipiden (meestal in celmembranen), wat leidt tot celbeschadiging. Kwantificering van lipideperoxidatie is essentieel om oxidatieve stress te beoordelen. Lipideperoxidatie vormt reactieve aldehyden zoals malondialdehyde (MDA) en 4-hydroxynonenal (4-HNE) als natuurlijke bijproducten. Malondialdehyde (MDA) en 4-hydroxynonenal (4-HNE) worden vaak gebruikt als markers van lipideperoxidatie en om te testen op oxidatieve schade/oxidatieve stress.
